    Political participation: the vocational motivations of Labour party employees

    Party employees are an under-researched group in political science. This article begins to address this oversight by examining Labour Party employees using new quantitative and qualitative data. It argues that party employment should be regarded as a form of political participation and as a consequence, existing models of political participation can be utilised to help explain why people work for political parties. After testing these propositions, the article concludes that existing models are indeed helpful in explaining the motivations for party employment

    The self-efficacy and confidence of Initial Teacher Education (ITE) students in understanding the learning needs of children with autism: Findings from a focus group discussion

    This research explored influences on self-efficacy and confidence for ITE students when teaching children with autism. Self-efficacy, defined by Bandura (1997), is a person’s perception of their capacity to carry out a course of action which will enable their attainment. The language of confidence was adopted when considering how ITE students experience self-efficacy. Qualitative data from interviews and a focus group discussion were analysed adopting an interpretative approach. Initial thematic analysis revealed the perceived importance of first-hand experience to develop knowledge and confidence, echoing Bandura’s (2006) argument that self-efficacy is developed through mastery experiences. The interrelated influence of emotional competency was also identified as pervading the discourse (Bunăiaşu, 2018). Interpretative analysis of the group discussion identified an increase in the perceived confidence of participants. Findings therefore support Bandura’s claim that talking develops self-efficacy and confidence through vicarious and persuasive experiences (Bandura, 1997). The vicarious experience enabled through the focus group appears to have supported emotional competence. Consequently, confidence and self-efficacy appear to have increased, when reflecting on teaching children with autism. Further research is needed to explore whether such experiences in focus group discussion can potentially inform pedagogical approaches to develop self-efficacy, confidence and emotional competence

    Using a new nomex lycra elastic fabric and individualized garment engineering techniques, reverse gradient garments (RGG's) were designed, constructed, and tested for effectiveness as a countermeasure against cardiovascular deconditioning. By combining torso compensated positive pressure breathing with a distally diminishing gradient of counterpressure supplied by the elastic fabric on the limbs, the RGG acts to pool blood in the extremities of recumbent persons much as though they were standing erect in 1 g. The RGG stresses the vasculature in a fashion similar to that experienced by the normally active man, hence preventing or limiting the development of post weightlessness orthostatic intolerance and related conditions. Four male, college age subjects received daily treatments with the RGG during a 15 day bedrest study. Four additional subjects also underwent the bedrest, but received no treatments; they served as controls. The preliminary indication was that the RGG was somewhat effective in limiting the deconditioning process

    ATPase cycle and DNA unwinding kinetics of RecG helicase

    The superfamily 2 bacterial helicase, RecG, is a monomeric enzyme with a role in DNA repair by reversing stalled replication forks. The helicase must act specifically and rapidly to prevent replication fork collapse. We have shown that RecG binds tightly and rapidly to four-strand oligonucleotide junctions, which mimic a stalled replication fork. The helicase unwinds such DNA junctions with a step-size of approximately four bases per ATP hydrolyzed. To gain an insight into this mechanism, we used fluorescent stopped-flow and quenched-flow to measure individual steps within the ATPase cycle of RecG, when bound to a DNA junction. The fluorescent ATP analogue, mantATP, was used throughout to determine the rate limiting steps, effects due to DNA and the main states in the cycle. Measurements, when possible, were also performed with unlabeled ATP to confirm the mechanism. The data show that the chemical step of hydrolysis is the rate limiting step in the cycle and that this step is greatly accelerated by bound DNA. The ADP release rate is similar to the cleavage rate, so that bound ATP and ADP would be the main states during the ATP cycle. Evidence is provided that the main structural rearrangements, which bring about DNA unwinding, are linked to these states

    Synthetic biology is principally concerned with the rational design and engineering of biologically based parts, devices, or systems. However, biological systems are generally complex and unpredictable, and are therefore, intrinsically difficult to engineer. In order to address these fundamental challenges, synthetic biology is aiming to unify a body of knowledge from several foundational scientific fields, within the context of a set of engineering principles. This shift in perspective is enabling synthetic biologists to address complexity, such that robust biological systems can be designed, assembled, and tested as part of a biological design cycle. The design cycle takes a forward-design approach in which a biological system is specified, modeled, analyzed, assembled, and its functionality tested. At each stage of the design cycle, an expanding repertoire of tools is being developed. In this review, we highlight several of these tools in terms of their applications and benefits to the synthetic biology community
